Abstract: Abstract of thesis entitled "ADRENOMEDULLIN IN THE RAT REPRODUCTIVE SYSTEMS AND THE CHANGES OF THE GENE EXPRESSION OF ADRENOMEDULLIN AND ITS RECEPTOR COMPONENTS DURING AGEING" Submitted by Li, Yuk Yin for the degree of Doctor of Philosophy at The University of Hong Kong in January 2008 Adrenomedullin (AM) is a multifunctional peptide originally isolated from pheochromocytoma. AM is widely distributed in rat tissues including reproductive organs. Previous studies demonstrated several roles for AM in regulating the functions of reproductive systems but the roles of AM in steroidogenesis as well as the age-related changes in AM biosynthesis and gene expression of AM receptor components remain to be elucidated. In the rat testis, positive AM immunostaining was found in the Leydig cells and Sertoli cells. Gel filtration chromatography showed that most of the immunoreactive AM in testis was the rat AM precursor. Specific AM binding sites were present in the testis and at least some of these AM binding sites can bind CGRP as demonstrated by displacement experiments. In vitro studies demonstrated an inhibitory effect of AM on hCG-stimulated testosterone secretion via its specific receptors. In the rat ovary, positive AM immunostaining was localized in the granulosa and theca cells of the follicle and granulosa lutein cells of the corpus luteum. Most of the immunoreactive AM in ovary was the active rat AM at estrus and rat AM precursor at diestrus. Specific AM binding sites were found in the ovary. In vitro studies showed inhibitory effects of AM on FSH-induced oestradiol secretion in 2-day old follicles and hCG-induced progesterone secretion in corpora lutea. These inhibitory effects of AM were mediated by CGRP receptors in the follicles but mediated by AM specific receptors in the corpora lutea. In the cycling rats, ovarian AM and its mRNA levels were decreased at estrus. By semi-quantitative RT-PCR, the expression of receptor components, namely calcitonin receptor-like receptor (CRLR), receptor activity modifying protein 1 (RAMP1) and receptor coupling protein (RCP) were down-regulated at estrus, metestrus and/or diestrus in the uterus. In ovariectomized rats, mRNA levels of AM, RAMP1 and RAMP2 were reduced in the uterus. With regard to ageing, male rats aged 3, 12 and 20 months and female rats aged 3 and 20 months were used. There was an age-associated increase in AM peptide and mRNA expression in the testis and decrease in the ventral prostate, seminal vesicle and epididymis. The gene expression of CRLR and RCP was only reduced in the sex accessory glands. Testicular RAMP1 and RAMP3 expression was upregulated with age. RAMP1 expression was down-regulated in ventral prostate, whereas RAMP2 and RAMP3 expression was reduced in seminal vesicle. All 3 RAMPs were decreased in the epididymis with age. In the female reproductive organs, AM mRNA levels were diminished with age in the ovary, oviduct and uterus but AM peptide levels were only reduced in the ovary and oviduct. The gene expression of CRLR and RCP was decreased with age in the ovary, oviduct and uterus. For RAMP expression, only RAMP2 was reduced in ovary while RAMP3 decreased in uterus.
